The short version of lyophilized powder fits in a sentence. The long version — which is the one that helps — is below.
Reviewed 2025-10-15. Anything still debated is marked as such rather than presented as settled.
Lyophilized peptide arrives as a dry cake that should stay sealed until use. Reconstitution is generally performed with sterile water or a buffered solution, and the resulting liquid should be handled gently to limit mechanical stress. Repeated freeze-thaw cycles are widely described as harmful to short peptides, so dividing a reconstituted batch into single-use portions is a common practice. Laboratories also record the solvent, concentration, and date of preparation on the vial label to keep later measurements traceable.
Dry powder is commonly held at minus twenty degrees Celsius, with some suppliers recommending lower temperatures for long-term archival storage. Once dissolved, solutions are typically kept cold and protected from light, since aqueous peptide solutions can lose integrity through hydrolysis or oxidation over time. Stability data specific to this fragment are limited in the public literature, and much of the guidance comes from general peptide handling practice rather than from controlled degradation studies. Users therefore treat stated shelf lives as approximate rather than fixed.
The compound is most often distributed as a lyophilized powder, appearing white to off-white and forming a loose cake or fluffy solid. It is hygroscopic to some degree, so brief exposure to humid air can add water weight and complicate weighing. The peptide dissolves readily in water and in neutral aqueous buffers, and aqueous solubility is generally described as high, well above the concentrations used in typical assays. Some polar organic solvents are also usable, which matters when a concentrated stock is prepared before dilution into buffer.
Storage recommendations center on keeping the dry powder cold, dry, and dark. A freezer at -20 degrees Celsius or below is conventional, and desiccant is often included to limit moisture uptake. Once dissolved, the peptide is less stable, and solutions are typically kept frozen and thawed only once. Repeated freeze-thaw cycles are a common source of losses because they promote aggregation and adsorption to container surfaces. Working aliquots are therefore prepared in advance, and glass or low-binding plastic is usually preferred over ordinary laboratory plastic.
Identity and purity are assessed with a small set of standard techniques. Reverse-phase high-performance liquid chromatography gives a purity estimate from peak area, usually recorded at 214 or 220 nanometers, where the peptide bond absorbs. Mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidized species. Amino acid analysis or tandem mass spectrometry sequencing can verify the sequence itself. Additional quality attributes include water content, residual trifluoroacetic acid carried over from purification, and endotoxin where the material is intended for biological work.
| Property | Value | Notes |
|---|---|---|
| Molecular mass | Approximately 0.9 kDa | Depends on exact fragment sequence and counterion |
| Amino acid sequence | LKKTETQ (commonly cited) | Short actin-binding motif from thymosin beta-4 |
| Common salt form | Acetate salt | Trifluoroacetate also reported in research material |
| Reconstitution solvent | Sterile water or buffer | Gentle mixing; avoid vigorous agitation |
| Solution storage | -20 °C or lower | Aliquot to avoid repeated freeze-thaw cycles |
Detection in biological matrices generally relies on liquid chromatography coupled with tandem mass spectrometry, because the peptide lacks a convenient ultraviolet chromophore beyond the amide backbone. Immunoassays have been described, but antibodies raised against the fragment can cross-react with the full-length protein or with unrelated peptides, so findings usually require confirmation by a second technique. Sample preparation typically involves protein precipitation followed by solid-phase extraction. Reported detection windows depend on dose, route, matrix, and instrument sensitivity.
Lyophilised material is stable for extended periods when kept dry and cold, and suppliers typically recommend storage well below freezing. Once dissolved, the peptide is handled at refrigeration temperatures and used within a short period, because peptide bonds and the acetylated terminus can be affected by repeated freeze-thaw cycles, proteases, or extreme pH. Bacteriostatic water and saline are both described as solvents, although preservatives can interfere with some analytical workflows. Reconstituted solutions are inspected for particulates before use.
TB-500 is a synthetic peptide preparation marketed under a name derived from thymosin beta-4, a 43-residue actin-binding protein first isolated from thymus tissue. The full-length protein has a reported molecular mass near 4963 Da, while material sold as TB-500 is often described as a fragment containing the actin-binding motif LKKTETQ. Because suppliers use the name inconsistently, published sources sometimes refer to the same label as a fragment, a synthetic copy, or a related analog. This naming ambiguity complicates direct comparison of reports across studies.
Laboratory work on thymosin beta-4 describes binding to monomeric actin and effects on cell migration, angiogenesis, and inflammatory signaling in cultured cells. Animal models have examined skin, corneal, and cardiac repair after injury, with outcomes reported mainly in preclinical literature. Most of that evidence concerns the parent protein rather than preparations labelled TB-500, so extrapolation from animal findings to a specific commercial product remains uncertain. Whether the two behave identically in living systems has not been established in controlled human studies.
TB-500 is a short synthetic peptide sold under a trade name rather than a systematic chemical name. Suppliers usually describe it as a fragment of thymosin beta-4 and ship it as a lyophilised powder intended for laboratory use. Because the label is commercial, the exact sequence attributed to it is not fully consistent across catalogues, and some listings present a seven-residue peptide while others describe related fragments of similar length. It is not an approved medicine in any major jurisdiction, and it is handled as a research chemical.
Thymosin beta-4 itself is a natural peptide of 43 residues found in many cell types and body fluids. Its best-characterised function is binding and sequestering actin monomers, which influences cytoskeletal dynamics. The sequence most often associated with TB-500, LKKTETQ, corresponds to part of that actin-binding region. A different fragment, Ac-SDKP, is also derived from the same parent peptide and is studied in its own right, which is one reason discussions of thymosin fragments can become confusing. The two are structurally distinct and are not interchangeable.
TB-500 is a synthetic seven-amino-acid peptide with the sequence LKKTETQ, corresponding to residues 17 through 23 of the protein thymosin beta-4. The N-terminus is typically acetylated in the described form, giving a monoisotopic mass near 888.5 Da and an average mass of about 889 Da. The designation TB-500 is a catalogue label rather than a formal chemical name, and the same sequence appears in the literature under several alternative abbreviations. It is handled as a research reagent rather than a pharmaceutical product.
Thymosin beta-4 is a 43-residue actin-binding protein found in most mammalian cell types, where it participates in cytoskeletal regulation and cell migration. TB-500 represents only a short fragment of that protein and does not include the remaining residues. Whether the isolated fragment reproduces the full range of activities reported for the intact protein remains an open question. Researchers commonly treat the two as related but distinct entities when comparing results.
The number of unique protein species likely will increase by between 50,000 and 500,000 due to RNA splicing and proteolysis events, and when post-translational modification also are considered, the total number of unique human proteins is estimated to range in the low millions. In addition, the first promising attempts to decipher the proteome of animal tumors have recently been reported. This method was used as a functional method in Macrobrachium rosenbergii protein profiling.
== Involvement in the programmed cell death == In contrast with the animal caspases, that exist in the cytoplasm in a form of pre-synthesized precursors, the activation of the phytaspase occurs during its maturation. However, by the time that the phytaspase molecule activates, it becomes physically separated form the supposed intracellular targets by the cell membrane due to the secretion process. Following the programmed cell death triggers, the phytaspase “re-enters” the cell and acts in the water-soluble fraction, where, presumably, it functions to degrade essential components for the cell homeostasis.
== Accords == On February 19, 2015, representatives from the Malian government and the Coordination of Azawad Movements (CMA) agreed upon a new document, including the cessation of hostilities, as negotiations to end the broader war continued in Algiers. On March 1, a mediation agreement was proposed by Algeria, which was accepted by Mali and pro-government militias. The CMA asked for some time to consider the agreement, as there were no propositions for autonomy or federalism for northern Mali, which angered a large portion of Tuareg rebels. The CMA announced their refusal to sign the agreement on April 10. On April 27, the pro-government GATIA and Arab Movement of Azawad (MAA) captured Menaka, initiating new clashes between Tuareg rebels and the pro-government militias. On May 10, the CMA initialized the agreement, but did not sign the text. The peace agreement was signed on May 15 in Bamako by the Malian government, representatives of pro-government militias, and representatives from Algeria, Burkina Faso, Mauritania, Niger, Nigeria, Chad, the African Union, the United Nations, ECOWAS, the Organisation of Islamic Cooperation, the European Union, and France. No CMA representatives were present for the signing on May 15. That same day, fighting broke out near Menaka. Under pressure from international organizations, the CMA finally signed the agreement in Bamako on June 20. The CMA's representative was Sidi Brahim Ould Sidati, a leader of the MAA.

Topiramate, marketed as Topamax among other names, is an oral medication primarily prescribed for the treatment of epilepsy and the prophylaxis of migraines. For epilepsy, this includes treatment for generalized or focal seizures. It has also been used off-label for alcohol dependence and essential tremor. Common side effects include tingling, feeling tired, loss of appetite, abdominal pain, weight loss, and cognitive dysfunction, such as trouble concentrating. Serious side effects may include suicidal ideation, increased ammonia levels resulting in encephalopathy, and kidney stones. Topiramate can cause birth defects, including cleft lip and palate. Risks and benefits should be carefully discussed with the full treatment team. Topiramate is considered "probably compatible" with lactation and is not contraindicated for breastfeeding, though monitoring of the infant for diarrhea or poor weight gain may be considered. Its mechanism of action is unclear. Topiramate was first synthesized in a search for an oral hypoglycemic agent; however, it received its initial approval as an anticonvulsant in 1996. It is available as a generic medication. In 2023, it was the 71st most commonly prescribed medication in the United States, with more than 9 million prescriptions.
==== Distribution of fitness effects (DFE) ==== Attempts have been made to infer the distribution of fitness effects (DFE) using mutagenesis experiments and theoretical models applied to molecular sequence data. DFE, as used to determine the relative abundance of different types of mutations (i.e., strongly deleterious, nearly neutral or advantageous), is relevant to many evolutionary questions, such as the maintenance of genetic variation, the rate of genomic decay, the maintenance of outcrossing sexual reproduction as opposed to inbreeding and the evolution of sex and genetic recombination. DFE can also be tracked by tracking the skewness of the distribution of mutations with putatively severe effects as compared to the distribution of mutations with putatively mild or absent effect. In summary, the DFE plays an important role in predicting evolutionary dynamics. A variety of approaches have been used to study the DFE, including theoretical, experimental and analytical methods.
Sadat became increasingly wary of Libya's radical direction, and the September 1973 deadline for implementing the Federation passed with no action taken. After the 1969 coup, representatives of the Four Powers—France, the UK, the US, and the Soviet Union—were called to meet RCC representatives. The UK and the US quickly extended diplomatic recognition, hoping to secure their military bases and fearing further instability. Hoping to ingratiate themselves with Gaddafi, in 1970 the US informed him of at least one planned counter-coup. Such attempts to form a working relationship with the RCC failed; Gaddafi was determined to reassert national sovereignty and expunge what he described as foreign colonial and imperialist influences. His administration insisted that the US and the UK remove their military bases from Libya. The British left in March and the Americans in June 1970. Moving to reduce Italian influence, in October 1970 all Italian-owned assets were expropriated, and the 12,000-strong Italian community was expelled alongside the smaller community of Libyan Jews. The day became a national holiday known as "Vengeance Day". Italy complained that this was in contravention of the 1956 Italo-Libyan Treaty, although no UN sanctions came. Aiming to reduce NATO power in the Mediterranean, in 1971 Libya requested that Malta cease allowing NATO to use its land for a military base, in turn offering Malta foreign aid. Compromising, Malta's government continued allowing NATO to use the island, but only on the condition that NATO would not use it for attacks on Arab territory.

Proteins are capable of forming very tight but also only transient complexes. For example, ribonuclease inhibitor binds to ribonuclease A with a roughly 20 fM dissociation constant. Other proteins have evolved to bind specifically to unusual moieties on another protein, e.g., biotin groups (avidin), phosphorylated tyrosines (SH2 domains) or proline-rich segments (SH3 domains). Protein–protein interactions can be engineered to favor certain oligomerization states.

Dry lyophilized powder is usually kept frozen, desiccated, and out of direct light. Sealed vials are not opened until needed, because moisture uptake can degrade short peptides. Longer archival storage is often done at lower temperatures than routine working stock.
Reversed-phase liquid chromatography separates components and reports purity from peak area. Mass spectrometry confirms the molecular mass expected for the sequence. Additional approaches such as peptide mapping or amino acid analysis provide independent confirmation.
Reported percentages depend on the analytical method, the detection wavelength, and whether salts and water are counted. A value above ninety-five percent by chromatography does not by itself establish a correct sequence. Different suppliers also calculate purity against different reference standards.
Short transit at ambient temperature is generally tolerated, but long-term storage at room temperature is not recommended. Heat, moisture, and light all accelerate degradation. Cold, dry, dark storage is the conventional choice.
